Kinetic Analysis of Dynamic PET with Targeted Probes –Theory and Application

The theoretical basis for a generic kinetics model for PET targeted probes will be presented.  This model accounts for the finite time needed by perfusion to deliver the probe to the tissue, bidirectional permeation of the blood-tissue barrier and the binding to and disassociation from the target. Computer simulation was used to study the accuracy of estimated model parameters using iterative optimization from probe uptake vs time curves with typical signal-to-noise ratio encountered in PET studies. Preliminary experience of using machine learning to estimate the model parameters will be presented.  Applications of the model to lung and pancreatic cancer will also be discussed.
